Mutant/mutation
The mutant expresses GFP under the control of the constitutive promoter of the elongation factor 1 alpha (eef1a) gene of P. berghei.

Phenotype
GFP is expressed in all blood stages and expression of the transgene has no influence on parasite viability and infectivity.

Additional information
Transgenic parasites develop to blood stages with no apparent defects and form gametocytes that exflagellate and can infect mosquitoes.
The DNA construct that has been used to introduce the gfp gene into the genome contains an incomplete copy of the P. berghei d-type ssu-rrna gene unit as a target region for integration into the genome of P. chabaudi. This construct has been used to introduce gfp in the genome of P. berghei (see RMgm-5).

This construct has been used to introduce gfp in the genome of P. berghei (see RMgm-5). Blast analysis of the P. chabaudi genome showed that there is highest identity (94%) between the P. berghei d-ssu-rrna target region and P. chabaudi ssu-rrna sequences on genome contig827, indicating that these are orthologous loci.
The gfp gene has been integrated into the genome by single cross-over integration. Therefore the possibility exists of reversion to the wild-type genotype by removal of the integrated DNA construct.
